Binding check

The “Binding Check” template is found in the “Assay Development” section of the DI.Control software represents the fastest and easiest way to iteratively test and improve the quality of an ideal binding assay. For a Binding Check assay, the signals of Target and Complex are compared to yield a qualitative yes/no answer to whether there is an interaction and a quantification of how well it can be resolved with the selected assay parameters. 

 

Binding Check experiments typically use Area of Response for TRIC measurements and the amplitude and S/N for Spectral Shift as analysis methods. It is recommended first to perform a Binding Check experiment to establish any given binding assay and then perform a Binding Affinity experiment for quantification of the affinity.

 

How to do a binding check:

One would create a new experiment after preparing the Target alone (Reference) and the target with the positive control (Positive Control). In the main menu, select “Binding Check” from the “Assay Development” menu.

 

If you only click with the mouse on the first well that you want to begin the experiment, the system automatically selects the 6 horizontal wells to the left and determines an equal number of each category (A1…A6, i.e.). If one wants to choose other orientations or a different number of assay points (4-6, etc.), keep the mouse still clicked and move in the desired direction and the number of wells one desires. Once the desired wells are selected, release the click button.
