What Does the Optical Unfolding Analysis App Do?

The Optical Unfolding Analysis application on NanoTemper Hub ingests raw Optical Unfolding data from Dianthus α, processes the unfolding curves, and delivers structured analytical outputs tailored to the following applications:

  • Protein quality assessment — for verifying that your protein is properly folded and stable before starting binding experiments

  • Binding validation — for confirming ligand binding by detecting ligand-induced stabilization, orthogonal to affinity measurements

  • Identifying problematic compounds — for flagging aggregators and auto-fluorescent compounds that may produce artifacts in your assay

  • Characterizing multi-component systems — for studying complex assemblies such as binary and ternary complexes, including PROTAC-mediated interactions
