Referencing

n Dianthus Single-Dose Screening experiments, the binding of ligands is evaluated by comparing the signal of the Ligand to the reference measurements. The processing of reference data for hit identification is referred to as referencing. Naturally, it is essential that the reference measurements are assigned correctly to the respective Ligand wells. In DI.Screening Analysis software, there are multiple options for this purpose that can be selected from a drop-down menu in the Data-tab of the software:

 

Referencing per Referencing ID:

The Referencing ID is an identifier that can be freely assigned to reference, ligand, and positive control wells in DI.Control software. It is used to define groups of wells as reference groups. If this option is selected, DI.Screening Analysis software will reference all data to the references that are in the same referencing group. This even applies to multiple plates.

 

Referencing per Row:

Ligand and positive control values in one row of the Dianthus plate are referenced to the reference wells within the same row.

 

Referencing per Column:

Ligand and positive control values in one column of the Dianthus plate are referenced to the reference wells within the same column.

 

Referencing per Plate:

All ligand and positive control values on a Dianthus plate are referenced to the reference wells of the same plate.

 

Referencing per Assay:

All ligand and positive control values of an assay are referenced in all reference wells of the same assay.
