Inconclusive

In the DI.Screening Analysis software, Ligands measured in screenings are automatically categorized into different categories to differentiate between Hits/Binders from Non-Binders or ligands that cause artifacts, such as Aggregation of the Target molecule.

Inconclusive is a Ligand Category, which has several sub-categories and include the following:

  • Inhomogeneous Response: A ligand in a single-dose screening is assigned to this category if the signal of the replicates deviates too much from the mean value of the ligand.

  • Potential Hit: A ligand in a single-dose screening is assigned to this category if a ligand-dependent decrease in Initial Fluorescence (i.e., Quenching) is observed. In such a case only a Specificity Test can reveal if this ligand is a true hit.

  • Not Reproducible: A ligand in a single-dose screening is assigned to this category if one of the Quality Checks returns different results for the measured replicates (e.g. one replicate showed aggregation whereas the other replicate did not).

  • Scan Anomaly: A well is assigned this category if the Well-Scan Quality checks show an anomaly in one of the Well Scans.

The ligand category inconclusive should not be confused with inconclusive results of Quality Checks.
