Measurement setup: size analysis

Size Analysis allows dynamic light scattering (DLS) measurements at a single temperature. This is why DLS is always set to ON for this measurement type. Carry out the following steps before starting a measurement in PR.Panta Control:

  • Choose between High-Sensitivity mode and Quick mode by setting the High-Sensitivity switch to ON or OFF, respectively. Quick size analysis measurements are, as the name implies, much faster than high sensitivity size analysis measurements. This speed comes at the tradeoff of sensitivity, resolution, and precision. For a quick snapshot of samples present at high enough concentration, the Quick mode is a good starting point. For precise sizing results and difficult samples, select High-sensitivity mode
  • Quick mode: 5 DLS acquisitions per capillary, acquisition duration 500 ms
  • High sensitivity mode: 10 DLS acquisitions per capillary, acquisition duration 5000 ms
  • Set the DLS laser power. The value for the DLS laser power is automatically carried over from the last selected discovery scan. You can modify the laser power manually between 1-100%.
  • Set a measurement temperature.
  • Select a solvent. In the annotations table, select a solvent (along with its viscosity) for each capillary. The viscosity is required to calculate the hydrodynamic radius.
  • Add annotations. Additional table columns can be added via themanageAnnotations_100x22.png dialogue. Annotations and annotation columns can also be modified via the same dialogue in the data table after the measurement has been completed.
