DLS acquisition time
A DLS acquisition represents a single dynamic light scattering measurement recorded from a capillary. To acquire DLS data, the intensity fluctuations of the scattering signal of a sample are correlated for the duration of a DLS acquisition. In Prometheus Panta acquisitions are either 5000 ms long in High-sensitivity mode of a size analysis measurement or 500 ms long in Quick mode of a size analysis measurement, as well as in thermal unfolding measurements.
Longer acquisition durations generally provide better data quality, but the short acquisition time of 500 ms has the advantage that it still gives good quality data while increasing acquisition speed. This is particularly important to achieve sufficient datapoint density in thermal unfolding measurements with 48 capillaries and linear thermal ramps. Each acquisition is evaluated separately and its result, the autocorrelation function, along with fitting results and quality parameters, can be accessed via the acquisition details tab of a measurement.
